Definition assigning/locating of a specific gene to particular region of a chromosome and determining the location of and relative distances between genes on the chromosome Reference National Center for Biotechnology Information, U.S. National Library of Medicine. Genome Mapping, http://www.ncbi.nlm.nih.gov/probe/docs/applmapping/ [31.10.2014]
Comment There are three types of maps:- linkage maps [ IATE:2211198 ]- physical maps [ IATE:1196421 ]- chromosome maps [ IATE:1625057 ]Some authors consider that genetic mapping is based on the use of genetic techniques to construct maps showing the positions of genes and other sequence features on a genome and therefore it constructs only linkage maps, and therefore they distinguish genetic mapping from physical mapping where molecular biology techniques to examine DNA molecules directly in order to construct maps showing the positions of sequence features, including genes.See also:- Brown TA. Definition map based on the direct analysis of DNA where physical distances between and within loci are measured in basepairs (bp), kilobasepairs (kb) or megabasepairs (mb) Reference Lee M. Silver. Mouse Genetics. Concepts and Applications. 7.1 Genetic maps come in various forms. Mouse Genome Informatics, http://www.informatics.jax.org/silver/frames/frame7-1.shtml [31.10.2014]
Comment Physical maps are arbitrarily divided into short range and long range. Short range mapping is commonly pursued over distances ranging up to 30 kb.Direct long-range physical mapping can be accomplished over megabase-sized regions with the use of rare-cutting restriction enzymes together with various methods of gel electrophoresis referred to generically as pulsed field gel electrophoresis or PFGE, which allow the separation and sizing of DNA fragments of 6 mb or more in length.Long-range mapping can also be performed with clones obtained from large insert genomic libraries such as those based on the yeast artificial chromosome (YAC) cloning vectors, since regions within these clones can be readily isolated for further analysis.
