Sainmhíniú method for the detection of a specific DNA sequence in DNA samples which combines transfer of electrophoretically separated DNA fragments to nitrocellulose paper for subsequent hybridisation with radioactive RNA or DNA probes Tagairt COM-Terminology Coordination, based on: Arizona State University. School of Life Sciences. Ask a Biologist. Mama Ji's Molecular Kitchen. 'Southern blot' (19.4.2021)
Nóta The idea for this approach arose from the technique developed by Ed Southern for analysing DNA fragments separated on agarose gels by capillary transfer to a nitrocellulose membrane where they could easily be probed by RNA or cDNA (Southern, 1975). The method of transfer is termed 'blotting' since the pattern of bands on the nitrocellulose membrane is an exact replica of the pattern in the original gel and therefore the technique for analysis of DNA became known as the 'Southern blotting' method. By a somewhat dubious geographical analogy, analysis of RNA molecules by a similar technique became known as 'Northern blotting' and transfer of proteins to membranes is sometimes called 'Western blotting'.
