Comment Cos sites are all that is necessary for packaging DNA into phage particles. The lambda DNA forms long concatamers, each lambda genome being joined to the next one through the cos site. The packaging enzymes chop the concatamer up into lambda-sized pieces by recognizing two cos sites 35-45kb apart and cleaving this unit which is then packaged into a phage.
