Definition "method for the detection of a specific DNA sequence in DNA samples which combines transfer of electrophoretically separated DNA fragments to nitrocellulose paper for subsequent hybridisation with radioactive RNA or DNA probes" Reference "COM-Terminology Coordination, based on: Arizona State University. School of Life Sciences. Ask a Biologist. Mama Ji's Molecular Kitchen. 'Southern blot' (19.4.2021)"
Comment The idea for this approach arose from the technique developed by Ed Southern for analysing DNA fragments separated on agarose gels by capillary transfer to a nitrocellulose membrane where they could easily be probed by RNA or cDNA (Southern, 1975). The method of transfer is termed 'blotting' since the pattern of bands on the nitrocellulose membrane is an exact replica of the pattern in the original gel and therefore the technique for analysis of DNA became known as the 'Southern blotting' method. By a somewhat dubious geographical analogy, analysis of RNA molecules by a similar technique became known as 'Northern blotting' and transfer of proteins to membranes is sometimes called 'Western blotting'.

    DNA vaccine
    en
Definition vaccine consisting in 'naked plasmid DNA', i.e. DNA without any associated lipid, protein or carbohydrate, which can elicit an immune response to the plasmin coded antigen Reference "COM-Terminology Coordination, based on:(1) nature.com > subjects > DNA vaccines (12.6.2020)(2) Restifo NP, Ying H, Hwang L, Leitner WW. 'The promise of nucleic acid vaccines (22.6.2020)'. Gene Ther. 2000;7(2):89-92. doi:10.1038/sj.gt.3301117"
Comment A DNA vaccine uses a gene from a virus or bacteria to stimulate the immune system. When the DNA vaccine is administered to a patient, the machinery in their cells makes a viral or bacterial protein which their immune system recognises as being foreign to the body. Like any vaccine the immune system will then recognise the bacteria or virus in the future – hopefully preventing illness.
