Definition A two-step technique for detecting antibodies reactive against cellular antigens. The unmodified serum is incubated with fixed cells containing the antigen. The specific antibody, if present, attaches to antigen sites. The presence of the attached antibody is detected by adding fluorochrome-labelled antiglobulin serum, which attaches to the previously fixed antibody and produces a marker visible by fluorescent microscopic examination. Definition fluorescent antibody test 1 in which unlabelled immunoglobulin (antibody) is added to tissue and combines with specific antigen, after which the antigen-antibody complex may be labelled with fluorescein-conjugated anti-immunoglobulin antibody, the resulting triple complex then being located by fluorescence microscopy 1 fluorescent antibody test [ IATE:1245167 ] Reference "indirect fluorescent antibody test." Comment Direct IF [ IATE:3576849 ] uses a single antibody directed against the target of interest. The primary antibody is directly conjugated to a fluorophore [ IATE:1910159 ]. Indirect IF uses two antibodies. The primary antibody is unconjugated and a fluorophore-conjugated secondary antibody directed against the primary antibody is used for detection. Reference: Abcam.
