- Comment In brief, SSBR consists of 1) strand break detection; 2) removal of 5′- or 3′-terminal blocking group; 3) gap-filling repair synthesis; and 4) nick-sealing by a DNA ligase. As reviewed herein, two proteins key to orchestrating SSBR are poly(ADP-ribose) polymerase 1 (PARP1) and X-ray cross-complementing protein 1 (XRCC1).
