Definition "mechanism that repairs normal and abnormal single strand breaks that arise either from reactions with DNA-damaging agents (endogenous or exogenous) or as intermediates in certain enzymatic events" Reference "COM-Terminology Coordination, based on: Abbotts, R., Wilson, D. M. 3rd., 'Coordination of DNA single strand break repair' (2.3.2021), Free Radical Biology and Medicine 2017; 107:228-244, doi:10.1016/j.freeradbiomed.2016.11.039"
Comment In brief, SSBR consists of 1) strand break detection; 2) removal of 5′- or 3′-terminal blocking group; 3) gap-filling repair synthesis; and 4) nick-sealing by a DNA ligase. As reviewed herein, two proteins key to orchestrating SSBR are poly(ADP-ribose) polymerase 1 (PARP1) and X-ray cross-complementing protein 1 (XRCC1).
