Definition "method of staining used to distinguish and classify bacterial species into two large groups by coloring these cells red (gram-negative) or violet (gram-positive)" Reference "COM-Terminology Coordination, based on: Monica Z. Bruckner, 'Gram Staining' (11.2.2020), Carleton College"
Comment Gram positive bacteria stain violet due to the presence of a thick layer of peptidoglycan in their cell walls, which retains the crystal violet these cells are stained with. Alternatively, Gram negative bacteria stain red, which is attributed to a thinner peptidoglycan wall, which does not retain the crystal violet during the decoloring process.

Definition immunohistochemical method used in diagnostics and research, involving the use of three antibodies:- a primary antibody binds to the antigen in tissue;- a secondary antibody reacts with the primary antibody;- a highly stable horseradish peroxidase-immunoglobulin complex acts as an antigen and reacts with the secondary antibody Reference "IHC World Life Science Products & Services > Knowledge Center > Immunohistochemistry Introduction, Introduction to Immunohistochemistry, http://www.ihcworld.com/_intro/ihc-methods.htm [13.6.2012]"
Comment An improvement on the indirect method of immunohistochemical staining. Has a very high sensitivity and allows for a high dilution of the primary antibody, eliminating many unwanted antibodies and reducing non-specific background staining.
