    Comment There are currently two different types of synthetic RNA vaccines: conventional mRNA and self-amplifying RNA (saRNA) (Fig. 1). The incorporation of chemically modified nucleotides, sequence optimization, and different purification strategies improve efficiency of mRNA translation and reduce intrinsic immunogenic properties [7]. However, antigen expression is proportional to the number of conventional mRNA transcripts successfully delivered during vaccination. Achieving adequate expression for protection or immunomodulation may thus require large doses or repeat administrations. saRNA vaccines, which are genetically engineered replicons derived from self-replicating single-stranded RNA viruses [8, 9], address this limitation.
