    Nóta Le gel de polyacrylamide est constitué d'acrylamide qui est l'unité de base et de bisacrylamide qui est l'agent portant. En faisant varier les taux de ces 2 substances on obtient différents maillages et donc différentes densités de gel. La polymérisation est réalisée grâce à l'ajout de 2 réactifs: le TEMED (N,N,N',N' tetraméthyl-èthylènediamine) et l'ammonium persulfate qui deviennent des anions hyperactifs en présence de lumière. Plus le pourcentage d’acrylamide est élevé, plus la densité des chaînes est élevée et les mailles du réseau sont serrées. Par conséquent, plus le pourcentage d’acrylamide est élevé, moins les molécules volumineuses peuvent migrer. Donc, plus que la masse moléculaire du composé est élevée, plus que la migration est lente.

    Nóta Pulsed-field gel electrophoresis (PFGE) is a highly discriminative molecular typing technique that is used in epidemiological studies worldwide. PFGE is based upon the variable migration of large DNA restriction fragments in an electrical field of alternating polarity. By comparing the fingerprints of any two isolates, one can investigate if they belong to the same strain (i.e. the two isolates are clonal) or if they are genetically unrelated. The implementation of PFGE in international surveillance networks, such as PulseNet, and the availability of standardized protocols for important foodborne pathogens (E. coli, Listeria, Campylobacter, …) have facilitated the adoption of this technique. Although more recent typing methods are available, PFGE is still the gold standard in many national and international surveillance programs.
