    Nóta Human embryonic stem cells (hESCs) are generated by transferring cells from a preimplantation-stage embryo into a plastic laboratory culture dish that contains a nutrient broth known as culture medium. The cells divide and spread over the surface of the dish. The inner surface of the culture dish is typically coated with mouse embryonic skin cells that have been treated so they will not divide. This coating layer of cells is called a feeder layer. The mouse cells in the bottom of the culture dish provide the cells a sticky surface to which they can attach. Also, the feeder cells release nutrients into the culture medium. Researchers have devised ways to grow embryonic stem cells without mouse feeder cells.     Nóta Such cells vary in their differentiation capacity, but it is usually limited to cell types in the organ of origin. Through in vivo and in vitro manipulations, TSCs have been identified in a variety of tissues (e.g. bone marrow, central nervous system, the epidermis, intestinal epithelium, skeletal muscle, the germ line, and shoot and root apical meristems), where they serve to replace cells that die, are lost due to injury, or are continually depleted during the life of the organism. TSCs differ from ESCs in that during normal development TSCs are more limited in their in vivo developmental potential, and their cell division gives rise to one daughter cell that acts as a stem cell and another that produces differentiated progeny. The stem cell state and the developmental capacity of the daughter cells is influenced by signals from the surrounding environment, and TSCs show plasticity in choosing their course of differentiation when their microenvironment is altered.     Nóta Stem cells are the very early cells that can develop into almost all other types of cell and tissue. They occur in the early (five-day) embryo when it is a tiny ball of about 100 cells before it implants in the uterus (embryonic stem cells or "ES cells"). They also occur in significant numbers in some tissues in the developing fetus and in umbilical cord blood at birth. They can also be found in some adult tissue, e.g. bone marrow, but they can be difficult to isolate, being present in very small numbers.Because of their ability to differentiate into so many different cells and tissues, stem cells hold out exciting prospects for the development of new cell-based therapies. They are already used for some treatments of cancers (bone marrow stem cells) and in early clinical trials in Parkinson's disease (fetal stem cells).
