Definition "pluripotent stem cell [ IATE:1077058 ] derived from an early stage human embryo, up to and including the blastocyst stage, that is capable of dividing without differentiating for a prolonged period in culture, and is known to develop into cells and tissues of the three primary germ layers" Reference "COM-EN, based on:'Human embryonic stem cell (hESC)' in 'Glossary', US National Institutes of Health Stem Cell Information, http://stemcells.nih.gov/StemCells/Templates/StemCellContentPage.aspx?NRMODE=Published&NRNODEGUID=%7b3C35BAB6-0FE6-4C4E-95F2-2CB61B58D96D%7d&NRORIGINALURL=%2finfo%2fglossary%2easp&NRCACHEHINT=NoModifyGuest#hesc [3.8.2011]"
Comment "Human embryonic stem cells (hESCs) are generated by transferring cells from a preimplantation-stage embryo into a plastic laboratory culture dish that contains a nutrient broth known as culture medium. The cells divide and spread over the surface of the dish. The inner surface of the culture dish is typically coated with mouse embryonic skin cells that have been treated so they will not divide. This coating layer of cells is called a feeder layer. The mouse cells in the bottom of the culture dish provide the cells a sticky surface to which they can attach. Also, the feeder cells release nutrients into the culture medium. Researchers have devised ways to grow embryonic stem cells without mouse feeder cells. This is a significant scientific advance because of the risk that viruses or other macromolecules in the mouse cells may be transmitted to the human cells.Reference: 'III. What are embryonic stem cells?' in 'Stem Cell Basics', US National Institutes of Health Stem Cell Information, http://stemcells.nih.gov/info/basics/basics3.asp [3.8.2011]See also:- stem cell [ IATE:1077058 ]- embryonic stem cell [ IATE:1036294 ]"

Definition "any pluripotent cell found in bone marrow that can differentiate into a variety of cell types found in the skeletal tissues, such as bone cells (osteoblasts), cartilage cells (chondrocytes), and fat cells (adipocytes)" Reference "COM-EN, based on:EuroStemCell. 'Mesenchymal stem cells: the 'other' bone marrow stem cells' (20.5.2020)"
Comment "1) These are not to be confused with haematopoietic (blood) stem cells that are also found in bone marrow and make our blood.2) The International Society for Cell & Gene Therapy (ISCT®) Mesenchymal Stromal Cell (ISCT MSC) committee issued a position paper in 2005 clarifying that the term mesenchymal stem cell is not equivalent or interchangeable with mesenchymal stromal cell (MSC). The former refers to a stem cell population with demonstrable progenitor cell functionality of self-renewal and differentiation, whereas the latter refers to a bulk population with notable secretory, immunomodulatory and homing properties. The ISCT's MSC committee further issued a minimal criteria to define multipotent MSCs as being plastic adherent, expressing CD73, CD90 and CD105, lacking the expression of hematopoietic and endothelial markers CD11b, CD14, CD19, CD34, CD45, CD79a and HLA-DR and capable of in vitro differentiation into adipocyte, chondrocyte and osteoblast lineages"

Definition "vaccine-related poliovirus which has a high level of VP1 sequence divergence from their parental OPV strains (>1% [types 1 and 3] or >0.6% [type 2]) indicating prolonged replication (or transmission) of the vaccine virus" Reference COM-Terminology Coordination, based on:
Comment -'Wild polioviruses are distinct and are not genetically linked to vaccine polioviruses.VDPVs are classified into three categories: a) circulating VDPVs (cVDPV) b) immune-deficiency-related VDPV (iVDPV), c) ambiguous VDPV (aVDPV). VDPVs reported as ambiguous VDPVs might later qualify to be iVDPV or cVDPV. The aVDPVs commonly occur in countries of low rates of polio vaccination coverage and could signal cVDPV emergence and gaps in immunization and surveillance activities' 1 -'The terminology can be confusing, because in principle all clinical and environmental vaccine-related poliovirus isolates can be considered to be vaccine derived. However, the term “VDPV” is specifically reserved for isolates whose extent of sequence divergence from the parental OPV strains indicates prolonged replication or transmission. The definitions follow from the poliovirus molecular clock, which ticks at an overall rate in which approximately 1% of nucleotides undergo substitution per year. Thus, vaccine-related viruses that differ from the corresponding OPV strain by >1% of nucleotide positions are estimated to have replicated in 1 or more persons for at least 1 year after administration of an OPV dose. The demarcation for type 2 VDPVs (VDPV2s) was lowered to 0.6% to increase the sensitivity for early detection of cVDPV2 outbreaks. 2'

Comment In describing a cell strain, its specific features must be defined. The terms finite or continuous are to be used as prefixes if the status of the culture is known. If not, the term strain will suffice. In any published description of a cell strain, one must make every attempt to publish the characterization or history of the strain. If such has already been published, a reference to the original publication must be made. In obtaining a culture from another laboratory, the proper designation of the culture, as originally named and described, must be maintained and any deviations in cultivation from the original must be reported in any publication.
